环氧化酶2抑制剂联合阿昔洛韦抑制Ⅰ型单纯疱疹病毒复活的实验研究 被引量:2

Experimental study of suppression of reactivation of herpes simplex virus type 1 by cyclooxygenase 2 inhibitor with acyclovir

摘要 目的探讨选择性环氧化酶2(COX-2)抑制剂对Ⅰ型单纯疱疹病毒(HSV-1)复活的抑制作用,以及联合使用阿昔洛韦的协同效应。方法本文为实验研究。实验小鼠完全随机分为6组,其中5组选取HSV-1在三叉神经节潜伏的小鼠,A组同时使用氯诺昔康和阿昔洛韦;B和C组分别单独使用两药;D、E两组腹腔注入生理盐水;第6组F组为未感染病毒的正常小鼠。6组中除E组外均进行紫外线照射。将角膜擦拭液或三叉神经节组织和293指示细胞共同培养,观测指示细胞有无出现病理性变化以鉴别有无病毒复制。结果A、B、C3个用药组的角膜和三叉神经节中病毒复活率与对照D组相比明显降低(角膜ХA-D^2=36.88,Х^2B-D=22.43,Х^2C-D=20.32,P〈0.05;三叉神经节Х^2A-D=49.91,Х^2B-D=29.16,Х^2C-D=24.89,P〈0.05);但联合用药的A组与单独用药的B、C组对角膜病毒复活率的比较差异无统计学意义(Х^2A-B=2.75,Х^2A-C=3.66,0.05〈P〈0.1);对三叉神经节病毒复制比较差异有统计学意义(Х^2A-B=4.78,Х^2A-C=6.97,P〈0.05)。结论选择性COX-2抑制剂氯诺昔康能显著抑制紫外线诱导的HSV-1的复活,联合使用阿昔洛韦眼液无显著的协同效应。该类药可为预防单纯疱疹病毒性角膜炎的复发提供新方法。 Objective To study whether the cyclooxygenase 2 (COX-2) inhibitor can block the herpes virus reactivation and whether the combination of COX-2 inhibitor with acyelovir can enhance the inhibition of virus reactivation. Methods It was a experimental study. Mice were randomly divided into six groups. Five groups were HSV-1 infected mice, which included: group A, treated with lomoxieam and acyelovir; groups B and C, treated with lornoxieam or acyelovir, respectively; groups E and F were injected with saline as the untreated control groups. The sixth group was uninfected mice as the control group. All groups were undergone to reactivate the herpes virus by UV-B except group F. The shedding of the virus was determined by cultures of ocular swab or ganglion homogenates with indicator cells. Results The rates of corneas and ganglia containing the infectious virus in the groups A, B and C were significantly lower than those in the control group D, (cornea:Х^2A-D =36. 88,Х^2B-D =22. 43,Х^2C-D =20. 32, P〈0. 05, ganglia:Х^2A-D= 49.91 ,Х^2B-D =29. 2 16,Х^2C-D=24.89, P 〈0.05). Combined use of these two drugs in group A showed no significant statistical difference as compared with using them separately in the cornea culture ( Х^2A-B = 2. 75, Х^2A-C = 3.66,0.05 〈 P 〈 0. 1 ), but there was significant difference in trigeminal ganglia culture ( Х^2A-B = 4. 78, Х^2A-C = 6.97 ,P 〈 0. 05). Conclusions These experiments demonstrate that a selective COX-2 inhibitor can suppress UV-B-indueed herpes virus reactivation in the cornea and nervous system. A combination of aeyelovir does not significantly enhance the inhibition of virus reactivation by lornoxieam. These results provide a new method to prevent the recurrence of HSK.
