摘要
目的建立人血浆中辛伐他汀的HPLC测定方法。方法采用Thermo-C18柱为分析柱;流动相为乙腈:水(60:40,V/V);流速为1.0 ml/min;检测波长为237 nm;柱温:35℃。结果辛伐他汀、洛伐他汀与人血浆中的内源性物质均实现较好分离,在0.5-25.0 ng/ml的范围内线性良好,r=0.9997(n=5),回收率为93.06%,日内及日间RSD均小于3.01%,最低检测限为0.25ng/ml。结论本法取样量小,操作简便快速,适用于临床辛伐他汀的血药浓度监测。
Objective To establish a high - performance liquid chromatographic (HPLC) assay for the determination of simvastatin in human plasma. Methods Thermo - C18 column was used: the mobile phase consisted of acetonitrile -water (60:40, V/V), the flow rate was 1.0 ml/min and the detection wavelength was at 237 nm at 35℃ temperature. Results A linearity was obtained from 0. 5 to 25.0 ng/ml of simvastatin in plasma with a good correlation coefficient ( r = 0. 9997, n = 5). The mean recovery was 93.06%. The intra day and inter day RSDs were less than 3.01%, and detection limit was 0. 25 ng/ml. Conclusion The improved method is sensitive, specific and simple and the method is suitable for the pharmacokinetics monitor.
