高压氧综合治疗大鼠脑外伤受损脑组织变化的实验观察(论著) 被引量:8

Experimental observation of the effect of combined hyperbaric oxygen treatment on injured brain tissue after brain trauma in rats

摘要 目的:观察高压氧(HBO)综合治疗对脑外伤受损脑组织的影响。方法:选用Wistar雄性同月龄(4个月)大白鼠132只,体重350~380g,在无菌条件下制成脑外伤模型。于伤后第8、9、10天各处死2只,经病理检查确定已进入脑外伤后软化灶形成期,然后随机分为3个组:HBO组、HBO综合组和药物组,并分别经3~9个疗程治疗后进行各组相互及治疗前后比较,观察各组动物脑外伤软化灶及其附近组织的病理改变差异情况。结果:药物组脑外伤软化灶的体积明显大于HBO组和HBO综合组(P<0.001),而HBO综合组的软化灶体积又较HBO组为小(P<0.001)。HBO组及HBO综合组在分别治疗3个疗程与治疗9个疗程后,脑外伤软化灶的体积变化亦有显著性差异(P<0.001)。病理检查可见在有效治疗后,脑外伤软化灶有神经细胞新生、再生和组织修复现象。在治疗6、9个疗程后,HBO组及HBO综合组总的基本恢复率均明显优于药物组(P<0.001)。结论:HBO治疗对脑外伤脑受损组织具有促进细胞新生和脑组织修复的良好作用。 Aim: To observe the effect of combined hyperbaric oxygen (HBO) treatment on injuredbrain tissue after brain trauma. Methods:The brain trauma animal model was made by using 132 maleWistar xats with same age(4 months) and weight of 350~380 g. The preparation was done using aseptictechnique and 2 of them were killed on the 8th, 9th and loth days respectively after brain trauma;andjudging from the formation of cerebromalacia focus detemined by pathological examination,all the re-maining rats were randomly divided into simple HBO,combined HBO and drug treatment groups. Com-parisons were made in order to determine the differences of the pathological changes in the cerebromala-cia focus and its neighbouring tissues after 3~9 courses of respective treatments. Results: The volume ofcerebromalacia focus in the drug group was obviously larger than those in simple HBO and the combinedHBO groups (P<0.001),and those in the combined HBO was smaller than that of the simple HBOgroup (P<0.001). There were significant differences in the volume of malacia focus between simpleHBO and combined HBO groups after 3~9 courses of respective treatment (P<0. 001 ). Pathologically,neogenesis of neurocytes and sign of tissue repair following effective treatment in the malacia focus werefound. The total basic recovery rate of simple HBO and combined HBO groups were obviously higherthan that of the drug group. Conclusion:HBO therapy has good effects in promoting the neogenesis ofneurocytes and repair of brain tissue after trauma.
