鸡马立克氏病疫苗荧光抗体染色阳性细胞数与蚀斑数线性回归(FAC^+ PR)方法的建立及应用

摘要 为解决鸡马立克氏病(MD)疫苗,尤其是MD多价疫苗在生产中疫苗配制环节上的盲目性,本试验建立了MDCVI988/Rispens/B5疫苗荧光抗体染色阳性细胞数(FAC+/mL)与疫苗蚀斑数(PFU/mL)两个数量化指标的线性关系(FAC+PR)。统计学检验证明,在此双变量下所建立的线性回归能客观反映该疫苗FAC+细胞数与疫苗PFU数之间的相互关系。将FAC+PR方法运用于MD疫苗的实验室制备,结果理论PFU/mL与实际PFU/mL的平均误差仅为5.4%。由此可见,采用这种方法可有效、准确估计出疫苗毒液中的PFU含量,并可以此为标准进行疫苗的配制。 To solve the confecting blindness of Marek' s disease (MD) vaccine in MD vaccine production, especially for MD polyvalent vaccines, linear regression was established between fluorescent antibody staining positive cells (FAC^+/mL) and plaques forming unit (PFU/mL) in this experiment. It has been proved that the established linear regression could objectively reflect the relationship between MD vaccine FAC^+/mL with MD vaccine PFU/mL each other by statistical tests. The results showed that the error of theoretical PFU/mL numbers to compared actual average PFU/mL numbers was only 5.4% when the FAC^+ PR had been used in MD vaccine confecting in laboratory. This results shows that the FAC^+ PR could be effectively used and accurately estimated the PFU numbers of MD vaccines, and could be used as a standard method for MD vaccine confecting.
