摘要
目的:鹿瓜多肽对去卵巢大鼠骨密度及松质骨中TGF-β1表达的影响。方法:将30只雌性SD大鼠,随机分为假手术(SHAM)组、去势(OVX)组、去势+鹿瓜多肽(OVX+CCP)组。术后20周测各组大鼠骨密度(BMD),利用免疫组织化学法观察各组大鼠松质骨中TGF-β1表达。结果:①OVX组与SHAM组相比较,松质骨中TGF-β1阳性细胞灰度值升高(P<0.05),CCP治疗组较OVX组灰度值降低(P<0.05);②与SHAM组相比,OVX组BMD明显降低(P<0.01);CCP治疗组各部位BMD高于去势组(P<0.05);结论:鹿瓜多肽注射液可以增加去势大鼠松质骨中TGF-β1的表达,抑制和延缓骨质疏松的形成,对雌激素缺乏引起的骨质疏松有预防作用。
Objective:To discuss the influence of cervus and cucumis polypeptide (CCP) on the expression of TGF - β1 in cancellated bone and bone mineral density (BMD) in ovariectomized rats. Methods:Thirty female SD rats were divided into three groups:sham operation (SHAM)group, ovariectomized (OVX)group and therapeutic (OVX + CCP )group. Bilateral ovariectomies were performed in OVX group and OVX + CCP group. The ovariectomized rats in OVX + CCP group were treated with CCP injection 1 day after operation. Twenty weeks after operation, the BMD of three groups was measured, and SABC immunohistochemistry was used to detect the expression of TGF - β1 in vertebrae bone. Results:The grayscale of cancellated bone in OVX group was significantly higher than that in SHAM group and OVX + CCP group ( P 〈 0.05 ). Compared with SHAM group, the BMD in OVX group was decreased significantly(P 〈0.01), and the BMD in OVX + CCP group was markedly higher than that in OVX group (P 〈 0.05 ). Conclusion: The CCP could increase the expression of TGF - β1 in cancellated bone of ovariectomized rats and inhibit the occurrence of osteoporosis in ovariectomized rats.
