摘要
目的研究水蛭提取物对体外培养新生大鼠大脑皮层神经细胞缺氧性坏死的保护作用。方法采用MTT法观察水蛭提取物对体外缺氧培养的神经细胞坏死的影响。结果水蛭提取物在0.5-2mg/ml浓度范围有抗神经细胞缺氧性坏死作用,其神经细胞坏死率明显下降,较凋亡模型组有显著性差异(P〈0.05),且随着浓度增加其抗缺氧性坏死作用随之增大。结论水蛭提取物对体外培养神经细胞缺氧性坏死有明显的保护作用。
Objective To study the protective effect of leech extract on apoptosis of primary cultured neonate rat cerebral cortical neurons caused by hypoxia in vitro. Methods MTT staining was used to observe the influence of leech extract on the necrosis of neurons caused by hypoxia. Results Leech extract was able to resist the necrosis of the neurons caused by hypoxia when the concentration of leech extract was between 500 μg/mL and 2 mg/mL. It decreased the death rate of neurons dramatically, having significant difference compared with apoptosis model group (P〈0.05) . Conclusions Leech extract has obvious protective function to necrosis of rat cerebral cortical neurons caused by hypoxia.
