柔嫩艾美球虫免疫与鸡盲肠扁桃体TNF-α基因表达动态的关系 被引量:5

摘要 根据GenBank上鸡β-actin、TNF-α的基因序列设计引物,应用逆转录-聚合酶链式反应(RT-PCR)技术克隆获得了β-actin和TNF-α基因,采用β-actin为内参的半定量方法检测TNF-αmRNA在鸡柔嫩艾美球虫免疫前后不同时间的表达情况,以探讨TNF-α基因的表达动态与柔嫩艾美球虫免疫的关系。结果显示,TNF-α基因在两次免疫期间的表达量整体上呈现双峰模式,首免后第9 d达到一个高峰,二免后第7d达到另一个高峰。结果表明,TNF-α在抗球虫感染中有一定的作用。 According to chicken β-actin and TNF-α gene sequences in GenBank, primers were designed, and the gene sequences were amplified by reverse transcriptase-polymerase chain reaction (RT- PCR) from the cecal tonsil of chicken experimentally immunized with Eimeria tenella. The expression level of TNF-α mRNA at different time after immunization was detected by semi-quantitative RT-PCR with β-actin gene as internal reference to study the relationship between E. tenella immunization and the kinetic expression of TNF-α gene in chicken cecal tonsil. The results showed that the level of expression of TNF-α gene reached 1st peak on day 9 post-lst-immunization, and reached 2nd peak on day 7 post-boosterimmunization. It was concluded that TNF-α had some effect against E. tenella infection.
