摘要
根据普通绵羊的碱基序列和相关文献设计引物,用RT-PCR扩增并测定了卡拉库尔羊肿瘤坏死因子-α基因的cDNA序列。结果表明,TNF-αcDNA序列包含了一个705 bp的开放阅读框,通过GenBank中的Blastn序列比较分析,卡拉库尔羊的TNF-α基因与普通绵羊的同源性高达99.86%。这为将来制备卡拉库尔羊TNF-α基因探针及卡拉库尔羊分子免疫学的发展提供了理论依据。
The prime was designed according to the DNA sequence of the ovis and relevant literature, the sequence of the kalakul's TNF - α gene was amplified by RT - PCR and sequenced. The result showed that the cDNA sequence of TNF - α contains a 705 bp of ORF. The comparison analysis of the fragment sequence with ovis through hlastn of GenBank, the homologies of the nucleotide sequence with ovis were 99.86 %. This research provide the scientific theoretical foundation on producing method of probe of kalakulg TNF - αand development of Molecular Immunology.
