摘要
以尾巨桉(E.urophylla×grandis)DH3229、DH3327、T13和巨桉(E.grandis)A44个桉树品种的组培生根瓶苗为材料,研究了不同盐浓度胁迫对其新生根、新生梢生长量等形态指标和叶绿素a、叶绿素b和总叶绿素含量的动态变化的影响,并分析了叶绿素含量与丙二醛(MDA)、脯氨酸(Pro)含量的相关性。结果表明,盐对桉树生长的影响随着盐浓度的升高和盐胁迫时间的延长而增加;虽然低浓度的盐胁迫使叶绿素含量略为增加,但随着NaCl浓度的提高,叶片中的叶绿素a、b和总叶绿素均随着盐浓度的增加而呈下降的趋势。叶绿素a/b值并未呈现单一变化趋势。相关分析表明,桉树叶绿素与膜脂过氧化产物MDA含量呈负相关,说明盐胁迫下叶绿素的降解与膜脂过氧化存在密切的关系。DH3229、T13叶片叶绿素与脯胺酸含量呈正相关;而DH3327和巨桉A4则呈负相关,表明游离脯氨酸的积累,对叶绿素的合成与降解并未起到直接作用。不同桉树品种对盐胁迫的反应存在显著差异,其中DH3229抗盐性最强,在盐逆境下其叶绿素较稳定并具有较高的生物产量,有望在盐碱地推广种植。
The tissue cultured seedlings of 4 varieties of Eucalyptus: E. urophylla ×grandis DH3229, DH3327 and T13, and E. grandis root number, the shoot A4 were treated with various concentrations of NaCl (0,0.2 %,0.4 %,0.6 %,0.8 %). The growth, and the biomass were measured from the seedlings in response to salt stress. Meanwhile, the dynamics of chlorophyll a, chlorophyll b and total contents of chlorophyll were studied and the correlation of chlorophyll contents with MDA and proline were analyzed. The inhibition of the growth and biomass was enhanced when NaCl concentrations increased or salt stress was prolonged. However, the contents of chlorophyll a, chlorophyll b, and total chlorophyll slightly when NaCl concentrations further increased. Correlation increased under low NaCl concentrations, but decreased analysis with the chlorophyll content, suggesting that the decomposition is showed that the MDA was negatively correlated of chlorophyll is closely related to the process of lipid peroxidation under salt stress. The relation between the total chlorophyll content and proline level was positive in DH3229 and T13,but was negative in DH3327 and A4,indicating that the regulation between chlorophyll and proline is variety-dependent. There was significant difference in response to salt Eucalyptus varieties.DH3229 is most resistant to salt stress,displaying a higher biomass chlorophyll content under salt stress condition, as compared to other varieties.
