摘要
采用单因子实验和正交试验对嗜热侧孢霉(Thermophilic sporotrichum)固体发酵产纤维素酶条件进行了研究,并测定了CMC酶、滤纸酶和棉花酶活力,以CMC酶和FPA酶为主要参考指标,最适产酶条件为:麸皮∶甘蔗渣(1∶2)50 g,(NH4)2SO4 1.0 g,K2HPO4 0.1 g,MgSO4.7H2O 0.03 g,含水量60%,pH5.5,培养温度为45℃,250 mL三角瓶盛放培养料40 g,培养4 d,测得CMC酶、FPA酶和棉花酶活力分别为320.04 IU,38.65 IU,180.31 IU。
It was studied that the optimum conditions of solid fermentation of Thermophilic sporotrichum to produce cellulase through single factor experiments and orthogonal test and the activities of CMCase,FPAase and cotton lyase were measured.The activities of CMCase and FPAase were used as main reference indexs.The optimal conditions of producing enzymes were as follows: 50 g bran+baggasse(1∶2),1.0 g(NH4)2SO4,0.1 g K2HPO4,0.03 g MgSO4·7H2O,water amount of medium was 60 %,pH was 5.5,40 g medium was put in 250 mL triangle flask.When cultured 4 days under 45 ℃,the activities of CMCase,FPAase and cotton lyase reached 320.04 IU,38.65 IU,180.31 IU.
