摘要
本文用高效毛细管区带电泳柱端安培检测了抗组胺药物异丙嗪(PMZ)。讨论了缓冲溶液浓度,pH值对柱效的影响,为提高检测的重现性考察了电极的电化学处理条件。在优化的实验条件下,异丙嗪的线性范围为1×10-7~1×10-4mol/L,检测下限为3.5×10-8mol/L,分离并检测了尿样中的1×10-7mol/L的异丙嗪。
An antihistamine drug, promethazine, was separated by high performance capil- lary electrophoresis and detected with an end-column amperometric detector. In this experi- ment, a fused silica capillary of 25 μm-i. d. and 37 cm-lenghth as a separation capillary, and a 30 μm carbon fiber disk electrode as working electrode were used. According to the hy- drodynamic voltammograms, the detection potential was selected at 0. 95 V(vs. Ag/AgCl). The effects of buffer pH and concentration on column efficiency have been investigated. Un- der the optimum conditions (separation voltage, 9 kV 3 citrate-phosphate buffer, 30 mmol/L; pH= 6. 56), the detection limit of promethazine was obtained to be 3. 5 x 10-8 mol/L (S/N =3). 1x10-7 mol/L of promethazine in urine was separated and detected.
