摘要
目的研究大蒜素(allicin)对急性髓系白血病(AML)患者骨髓单个核细胞(BMMNC)的作用并探讨其机制。方法采用大蒜素在体外作用于AML患者BMMNC,经MTT法、TdT缺口末端标记(TUNEL)技术、RT-PCR等方法检测大蒜素对AML患者BMMNC的影响。结果不同浓度大蒜素对AML患者BMMNC有不同程度的生长抑制、诱导凋亡和抑制iNOSmRNA表达的作用。①MTT法检测大蒜素对细胞的增殖抑制作用,发现大蒜素在3个浓度范围内对细胞均有增殖抑制作用,并呈量效关系。②TUNEL检测见阳性细胞核深染,大蒜素处理组与对照组比较,凋亡细胞明显增多。③RT-PCR检测发现大蒜素能显著抑制iNOSmRNA表达。结论大蒜素可抑制AML患者BMMNC生长、诱导期凋亡及抑制iNOSmRNA表达。
OBJECTIVE To study the effect of allicin on bone marrow mononuclear cells(BMMNC) of the patients with acute myeloid leukemia (AML). METHODS BMMNC of AML patients were treated with allicin in vitro. MTT,TUNEL and RT-PCR were used to investigate the effects. RESULTS ( 1 ) Compared with the control group, the number of BMMNC of AML patients in allicin group decreased obviously ; and has a relationship between dosage and effection. (2) Positive cells of hyperchromism were observed with TUNEL. Apototic cells increased significantly in allicin group, compared with the control group. (3)Compared with the control group ,the expression of iNOS mRNA in allicin group decreased obviously;and has a relationship between dosage and effection. CONCLUSION Allicin can inhibit the proliferation, induce the apoptosis and suppress the iNOS mRNA expression in BMMNC of AML patients in vitro.
