摘要
目的:选用生物相容性好的明胶作为褐藻多糖硫酸酯的固定载体,制备褐藻多糖硫酸酯与明胶的复合材料,采用体外凝血试验评估其抗凝血性能。方法:实验于2003-12/2006-12在北京理工大学材料中心实验室、中国科学院北京化学研究所光谱实验室完成。①实验材料:褐藻多糖硫酸酯从市售海带中采用溶剂萃取法制备,纯度可达40%。实验血浆由北京北太平庄血站提供。②将质量浓度分别为100,200,300mg/L的褐藻多糖硫酸酯0.1mL滴加到盛有0.1mL明胶的醋酸溶液(w=0.01)比色杯中,反应完全后,采用0.05%戊二醛固定,干燥成膜后用PBS缓冲溶液冲洗,然后真空干燥72h得到3种不同复合材料。采用EQINOX55型红外光谱仪(美国NICOLET公司)进行结构表征。③采用体外抗凝血时间实验评价复合材料的抗凝血性,包括部分凝血活酶时间、凝血酶原时间、凝血酶时间测定。结果:①复合材料的红外光谱图在877cm-1出现新吸收峰。②实验血浆样品的部分凝血活酶时间、凝血酶时间、凝血酶原时间分别为46.1s,14.0s,12.2s,在正常人血浆的部分凝血活酶时间、凝血酶时间、凝血酶原时间范围内。200mg/L复合材料能显著延长部分凝血活酶时间和凝血酶原时间(P<0.05),300mg/L复合材料能显著延长凝血酶时间(P<0.05)。结论:褐藻多糖硫酸酯-明胶复合材料具有较好的抗凝血活性。
AIM: To prepare a blend material of Fucoidan and gelatin, which possesses good biocompatibility, and investigate its anticoagulant activity by in vitro coagulation test. METHODS: The experiment was conducted at the Research Center of Material Science, Beijing University of Technology and the Spectrum Laboratory, Beijing Institute of Chemistry, China Academy of Sciences from December 2003 to December 2006. (1)Fucoidan was prepared by market kelp using solvent extraction method, with the purity of 40%. Blood plasma was offered by Beijing North Taipingzhuang Blood Station. (2)0.1 mL Fucoidan solution at different concentrations (100, 200, 300 mg/L) was added to 0.1 mL gelatin dissolved in acetic acid solution (w=0.01) in a covette. And 0.05% glutaraldehyde solution was added for fixation when the reaction completed. Three blend materials were formed after dried. The blend material was washed with PBS buffer solution and then dried at vacuum for 72 hours. The structural characteristics was examined through a infrared spectrometer (NICOLET, America). (3)The anticoagulant activity of blend material was assessed by in vitro coagulation time test including activated partial thromboplastin time (APTT), activated thromboplastin time (PT) and thrombin time (TT). RESULTS: (1)The blend material showed absorption bands at 877 cm^-1 of spectrum. (2)The APTT, TT, PT of blend material were 46.1 seconds, 14.0 seconds and 12.2 seconds, respectively, which were within those indexes of normal people's plasma. 200 mg/L blend material could remarkably prolong APTT and PT (P 〈 0.05), while 300 mg/L blend material increased TT (P 〈 0.05). CONCLUSION: The Fucoidan/gelatin blend material possesses good anticoagulant activities.
