摘要
本研究比较了鼠尾藻醇提取物(ST_1,ST_2,ST_3)体外抑制人食管癌细胞株(Ec-(109))的生理活性。结果表明,浓度为100mg/mL的不同提取物作用48h后均出现明显抑制,细胞大部分脱落,抑制率在75%—95%之问;当浓度为50mg/mL,抑制率保持在30%以上;25mg/mL以下浓度仍表现出显著效果。以5%浓度,445mg/kg ip连续10d对小鼠肉瘤S_(180)均有效果。 对提取物的生化研究发现,ST_3对荷瘤鼠血浆巯基(SH)含量有明显影响。ST_1和ST_3对荷瘤鼠血浆A/G比值有显著影响,结果提高了血浆内球蛋白浓度。ST_1对荷瘤鼠压积红细胞超氧化物歧化酶(SOD)有一定的影响,试验组SOD活性较对照低30%。ST_2可刺激人粒细胞超氧阴离子自由基(O_2)的释放,表明ST_2对粒细胞有激活作用。
We have formed a simple and convenient method on the basis of Ito's method in order to study the physiological activity of alcohol extracts (ST1,ST2 and ST3 ) from Sargassum thunbergii. A comparative study was made of the inhibition effect of the alcohol extracts on human esophageal cancer cell strain (109) in vitro. The results showed that esophgaeal cancer growth is obviously inhibited by all the three of alcohol extracts at the concentration of 100mg/mL. Most of the cancer cells dropped 48 hours after reaction, the inhibition rate ranging from 75% to 95%. The inhibition rate was still over 30% even when the concentration of each of the alcohol extracts was reduced to 50mg/mL. The inhibition effect of Sargassum thunbergii alcohol extracts on cells could still be seen even at the concentration of less than 25mg/mL.
Intraperitoneal injection of 5% ST1 3 solutions at 445mg/kg. d for 10days decreased the growth of S180 by 34. 55%( P >0. 05).55%( P <0. 05) and 50%( P <0. 01), respectively. The results of the experiments proved that ST2 and ST3 have a considerable inhibition effect on tumor. The extract ST3 has striking effect on the SH group in blood plasma of mice with cancer. Intraperitoneal injection of the extract ST3 for 10 successive days can reduce the SH group in blood plasma by 29. 5% to 35. 45% ( P<0. 01 ) The extracts ST1 and ST3 have a distinct effect on the blood plasma A/G ratio, increasting the globulin concentration of blood plasma by 22% to 26%. The extract STj has some effect on the superoxide dismutase of the compacted deposits of red cells,decreasing it by 30%.
Release of superoxide anion from human granulocytes stimulated by the extract ST2 was
also studied. Some amount of O2 could be released from granulocytes when ST2 or canavaline A (Con A) was applied alone. Pretreatment of granulocytes with low concentration ST2
potentiated O2 release from granulocytes stimulated by Con A,and the lag time of O2 release was reduced. The results showed that the granulocytes were activated by ST2. This phenomenon may be related to some action mechanism of ST2.
