摘要
目的研究过氧化物酶体增殖激活物受体γ(PPARγ)在皮肤鳞状细胞癌(CSCC)中的表达及其意义。方法用RT-PCR方法检测PPARγ的mRNA表达水平,用免疫组化方法检测PPARγ的蛋白表达水平。结果免疫组化研究结果表明:CSCC患者组PPARγ的表达强度显著低于正常对照组(x^2=6.09,P=0.048),且Ⅱ级CSCC患者组显著低于Ⅰ级CSCC患者组(x^2=8.40,P=0.15);RT-PCR研究结果表明:CSCC患者组PPARγmRNA的平均表达水平为0.29±0.12,对照组为0.74±0.22,患者组亦显著低于对照组(t=7.22,P<0.001)。结论CSCC患者皮损部位PPARγ的表达降低,在CSCC的发病机制中可能起一定的作用,有可能成为治疗CSCC的新靶点。
Objective To investigate the expression of peroxisome proliferators-actived receptoγ （PPARγ） in cutanous squamous cell carcinoma （CSCC） and to evaluate its possible significance. Methods The expression of PPARγ mRNA and protein was detected by RT-PCR and immunohistochemistry methods respectively in patients with CSCC and normal controls. Results The mRNA level of PPARγ in CSCC patients was significantly lower than that of normal controls. Immunohistochemical study showed that the expression of PPARγ protein was also markedly decreased in CSCC tissues than that of normal controls. Conclusion The expression of PPARγ is significantly decreased in CSCC patients than that of normal controls,it may play a role in the pathogenesis of this disease, and PPARγ may be a novel target to the treatment of CSCC.
