摘要
目的观察BALB/C小鼠腹腔荷瘤模型,经温热处理后其平均存活时间及温热后不同时间腹腔各主要脏器和肿瘤组织的光镜和电镜组织形态学变化。方法以B16黑色素瘤细胞经腹腔注入建立荷瘤鼠模型,小鼠腹部水浴加热41℃60min,处理后不同时间进行组织学和酶组织化学(琥酸脱氢酶,5-核苷酸酶)分析。结果经41℃温热,荷瘤鼠平均存活时间无明显变化(肿瘤对照组:59天;温热组:58天,P>0.05)。组织形态学变化:即时加热荷瘤鼠组与荷瘤鼠对照组相比无显著变化。荷瘤鼠加热一周后,肿瘤组织中心部位部分血管栓塞,可见肿瘤坏死灶。肝细胞空泡样变化,粗面内质网及线粒体堆积成块;脾红髓内可见较多含瘤细胞碎片的巨噬细胞;加热两周后,肿留组织出现不含黑色素颗粒的新鲜肿瘤细胞团,肝、脾、肾组织有不同程度的恢复。酶组织化学显示:各组琥酸脱氢酶,5-核苷酸酶含量无明显差异(P>0.05)。结论 41℃温热对荷黑色素瘤小鼠有一过性抑瘤作用,且对各正常脏器无明显影响。
Objective To examine, the morphological and histochemical alterations of melanoma and normal tissues in melanoma-bearing BALB/C mice treated with hyperthermia. Methods The mice bearing melanoma were given local heating in abdomen at 410C for 1 h. Treated melanoma and normal tissues were observed morphologically under light microscope and electron-microscope. Their succinate dehydrogettase and nucleotidase contents were assayed by histochemical method. Results Hyperthermia alone at 41℃ for one hour produced no enhanced viability of mice bearing melanoma. Morphologically there was no significant change for instant heating group as compared with the melanoma control group, but abput one week later after heating, thrombosis was observed at the center of the melanoma, and tumor necrosis also happened around it. In normal tissues, RER and mitochondria were accumulated in hepatocytes, and many phagocytes engulfing necrosis tumor fractions were observed within spleen red pulps. Two weeks later after heating, fresh tumor cells without melanin granules were ob- served, and normal tissues recovered to different extents. There was no histochemical changes, as SDH and 5'-AMPase con- tents in both groups had no significant difference after heating at 410C for 1 h. (PLO.05). Conclusion Heating at 41℃ for 1 h, had transient anti-tumor effects on mice bearing melanoma, but no effect on normal tissues.
