摘要
提出辣根过氧化物酶法分析左旋多巴,方法基于一定条件下左旋多巴对辣根过氧化物酶催化对氯苯酚-4-AAP偶联发色体系有干扰,干扰程度与左旋多巴的浓度有相关性。研究了干扰产生的原因和条件,建立了微量左旋多巴的测定方法,检出限可达 2.5 × 10-7mol/L。用于左旋多巴片的含量测定,结果令人满意。
A method was developed for the enzymatic assay of levodopa. The method is based on the interference to the coupling colorimetric system of p-chloropheno1ic acid and 4- aminoantipyrine catalyzed by horseradish peroxidase (HRP). The resulting change in ab- sorbance at maximum wavelength of coupling product is dependent on the concentration of levodopa. The mechanism of interference was studied preliminarily, and the optimum experi- mental conditions for the measurement were examined. The enzymatic method was used for measuring levodopa with a detection limit of 2. 5 ×107 mol/L. Determination of levodopa in tablet shows excellent correlation with the resu1ts of official methods of the same samples.
