养肝熄风方药对脑缺血大鼠神经干细胞增殖的影响 被引量:3

摘要 目的:观察养肝熄风方药对成年大鼠缺血脑损伤后神经干细胞增殖的疗效。方法:采用大鼠局灶性脑缺血模型,观察大鼠缺血损伤后,侧脑室下区(SVZ)的细胞增殖。结果:(1)养肝熄风组再灌后第7天脑梗死灶体积明显小于模型组(P<0.01);养肝熄风组大鼠在第7天、9天的神经功能缺损评分要明显好于模型组(P<0.05)。(2)和模型组相比,养肝熄风组大鼠SVZ巢蛋白阳性细胞表达于术后第7天显著增加(P<0.05),缺血损伤后第14、21天SVZ的神经干细胞增殖显著增加(P<0.01)。结论:(1)脑缺血损伤可激活SVZ神经干细胞,促使其增殖和表达。(2)养肝熄风方药能减轻急性大脑中动脉线栓法(MCAO)大鼠脑梗死灶体积,改善神经功能缺损。(3)养肝熄风方药可以促进MCAO大鼠SVZ神经干细胞的增殖。 Objective.To examine the effect of Yanggan Xifeng medicine (nourishing the liver to stop the wind) on proliferation of neural stem cells in rats with cerebral ischemia. Methods:To observe cell prolif-eration in SVZ area in rats with the setting of focal cerebral ischemia model. Results: (1) Compared with the model group, infarction volume in Yanggan Xifeng group was observed to reduce at day 7 after reper-fusion (P〈0.01) ; neuro-function deficit score was obviously better at day 7 and day 9 (P〈0.05). (2) Compared with the model group, Yanggan Xifeng group had a marked increase in SVZ positive nestin cell at day 7 ( P〈0.05), and a marked increase in SVZ cell proliferation at day 14 and day 21 after cerebral ischemia (P〈0.01). Conclusion: (1) Cerebral ischemia can activate SVZ neural stem cells, stimulating their proliferation and expression. (2) Yanggan Xifeng medicine can reduce infarction volume in the rats of MCAO (middle cerebral artery occlusion), improve the recovery of neural function deficit. (3) Yanggan Xifeng medicine can stimulate the proliferation of neural stem cells of MCAO rats.
