摘要
目的检测中药复方双黄连注射液对细菌酯多糖(LPS)诱导的血管内皮细胞核转录因子-κB(NF-κB)活化的抑制效应。方法用双黄连注射液与人脐静脉血管内皮细胞株(ECV-304)共同培养后,采用MTT法观察双黄连注射液对人脐静脉血管内皮细胞生长的作用,用Sandwich ELISA法及定量免疫细胞技术,检测双黄连注射液对细菌LPS刺激后的血管内皮细胞胞浆(IкB)及核内NF-κB含量变化的影响。结果较高浓度的双黄连注射液对血管内皮细胞的生长有抑制作用,其抑制强度与其浓度呈正相关;血管内皮细胞在LPS刺激下,胞浆内IκB蛋白的含量显著降低,核内NF-κB p65含量显著增高;双黄连注射液预处理后能显著抑制IкB的降解及NF-κB的表达,在亚细胞生长抑制浓度条件下,其抑制效应有统计学意义。结论双黄连注射液可负相调节细菌LPS介导的血管内皮细胞核转录因子-κB的活化。
OBJECTIVE To examine the effect of traditional Chinese drug compound Shuang Huanglian injection (SHLI) on the LPS - mediated NF - κB activation in human umbilical vein endothelial cells (HUVECs). METHODS SHLI and ECV -304 were co - cuhured. The effects of SHLI on the growth of HUVECs were observed by MTT and morphological method. The amounts of cytoplasm IκB and nuclear NF - κB in ECV - 304 cells with or without LPS treatment were determined by the quantitative immunocyte assay and Sandwich ELISA quantitative assay. RESULTS SHLI had some inhibitory effects on the growth of ECV304, and this effect was positively correlated with the concentration of SHLI. A significant increase in the nuclear NF - κB was detected in the LPS - stimulated ECV304 cells pretreated by SHLI. Coincidated with that, the IκB reduction of cytoplasm was determined in the LPS - stimulated ECV304 cells pretreated by SHLI. CONCLUSION SHLI can down - regulate LPS - mediated NF - κB activation in the human vascular endothelial cells.
