紫色甘薯的茎尖培养与脱毒 被引量:13

摘要 以紫色甘薯品系为试材,研究6-BA和NAA及基因型对紫色甘薯茎尖培养的影响,并采用硝酸纤维素膜—酶联免疫法(NCM-ELISA)和症状法对再生植株进行病毒学检测。结果表明,培养基中6-BA和NAA浓度和配比对紫色甘薯茎尖培养的愈伤组织、不定根和不定芽的诱导有明显影响。6-BA可促进不定芽的诱导,附加1mg/L6-BA的MS培养基为最佳诱芽培养基。基因型对诱芽率也有一定的影响,其中品系B3和B7的诱芽率可达50%。通过病毒学检测,获得12个紫色甘薯品系的脱毒苗,其平均脱毒率为94.7%。 In this paper,effects of 6-BA,NAA and genotypes on shoot tip culture in vitro were studied by using the strains of purple-colored sweet potato introduced as test material.The condition of virus infection in regeneration plantlets were detected by NCM-ELISA and visual estimate.The results showed that the concentration and combination of 6-BA and NAA in MS medium had obvious effects on induction of callus,adventitious root and adventitious bud from shoot tip explants of purple-colored sweet potato.6-BA could improve the induction of adventitious bud,and the MS medium containing 1 mg/L 6-BA was the best medium for adventitious bud.The genotypes of purple-colored sweet potato also affected the frequency of adventitious bud induction,and the frequency of adventitious bud induction in Strain B3 and B7 could reach 50 %.The virology detection showed that virus-free regeneration plantlets were obtained in 12 strains of purple-colored sweet potato,the average rate of virus elimination was 94.7% depend on different genotypes.
