摘要
甲状旁腺激素(PTH)目前已作为一种促骨合成药物,用于骨质疏松症的临床治疗。它所发挥的生物学效应取决于其作用模式和剂量,即间断小剂量使用能增加骨形成,而大剂量持续使用则加速骨吸收使骨量减少。PTH对骨代谢调节的确切机制尚未阐明,大量研究工作力图从各个层面寻找PTH治疗骨质疏松的效应模式及机制,以便使其更安全有效地应用于临床,本文对目前的研究进展作综述。
Parathyroid hormone (PTH) and its analogs have been used as bone anabolic agents for the treament of severe osteoporosis in clinic. PTH exerts biphasic effects on osteoblastie cells, depending on dose, duration of exposure. The anabolic actions of PTH are dependent on an intermittent regimen. On the contrary, continuous administration will result in bone loss through the increase in resorption. Its mechanisms of action on bone cells are unclear. This article reviews the progress in the researches on the effect of PTH and its analogs on osteoporosis.
