摘要
经过研究建立了盐析、离子交换层析和凝胶过滤层析的分离纯化程序,从地木耳(Nostoc commune)细胞中分离纯化出藻蓝蛋白的α和β两个亚基,其纯度(A615/A280)为5.7,最大吸收峰为615 nm,荧光发射峰为560 nm,α和β亚基的分子量分别为18.1 kD和19.1 kD。
The phycocyanin is a kind of blue natural protein with the characteristics of absoption spectra and fluorescence spectra, its basic structure is composed of subunit a and β. The successive procedures of fraction precipitation of ammonium sulfate, ion exchange and gel filtration chromatography were set up. Subunit a and β of phycocyanin from Nostoc commune was isolated and purified, the purity(A615/A280) reached 5. 7, the max absorption spectra was 615 nm and the fluorescence emission spectra was 560 nm. Molecular weight of subunit a and β were 18.1 kD and 19.1 kD respectively.
