摘要
以鱼腥藻 (Anabeana)为材料 ,利用超声波进行细胞破壁 ,采用正交试验 ,从中选出最优工艺条件 ,即原料重 (鲜重 ) 8倍体积的磷酸缓冲液 ,超声波功率 6 0 0W ,作用 9min后 ,静止 30min ,藻蓝蛋白释放完全 ,饱和度为 4 0 %的硫酸铵使藻蓝蛋白盐析的效果较好 ,紫外可见光谱、荧光光谱等测定表明 ,所提藻蓝蛋白分别在 6 5 1 6nm和 6 96 6nm处有明显特征吸收峰。
The Anabaena was used in the experiment to study the cellulate corpuscle l. The optimal conditions were presented using sonication to disrupt Anabaena. The experiment was designed by orthogonal method and the result was as follows. Add phosphoric acid buffer 20 mL,which is 8 time of the fresh Anabaena. Sonicate 9 mins using power of 600w,then the phycocyanin will be released after incubate the mixture for 30 min at room temperature. The phycocyanin was salted by the 40% saturation of ammonium sulphate. The sample was detected by the ultraviolet and the fluorescence methods and we found that the phycocyanin has maximum absorption peak at 651.6 nm while the allophycocyanin has maximum absorption peak at 696.6 nm.
