克隆蓝藻Calothrix sp.PCC7601 cpcB2A2基因的初步研究

摘要 从蓝藻Calothrixsp.PCC7601染色体DNA中分离出含藻蓝蛋白基因cpcB2A2的略3.8KbDNA片段,与质粒pUC18重组,构建成一种重组质粒pPC218-PCZ,转化E.coliJM109,获得了一系列克隆子.经斑点分子杂交和限制性内切核酸酶分析,筛选出初步认为含有cpcB2A2基因的克隆子. Biddulphia regia was collected from Xiamen coast in August 1987.Sexual process of this diatom was observed. Sex Differentiation occured only once a day that lasted 4 hours with the light induction. When the cell size reduced to the range of 26~62 μm in widths ,the sexual process set in ,There were 128 sperms in each spermatogonangum. The survival rate of sperms declined greatly 19 hours after they were formed. Each oogonium contained two eggs. Sperms and eggs were produced at the same time. The results indicated that propaly in this diatom sexualization took place separately in the cells of small size range and middle size range, and it involved two times of restoration of cell sze. The similarities and differences in sexual reproduction showed the relationship of species.
