大鼠创面愈合过程中表皮干细胞增殖分化与玉红膏的影响(英文) 被引量:5

摘要 背景: 表皮干细胞在创伤修复中具有重要作用,从诱导干细胞角度来阐释中药加速受损皮肤再生的机制值得研究。目的: 观察玉红膏对大鼠创伤愈合过程中皮肤组织中表皮干细胞增殖、分化的影响。设计: 随机对照动物实验。单位: 中国中医科学院望京医院药理室。材料: 实验于 2006- 07/09 在中国中医科学院望京医院药理室完成。选用114 只清洁级健康 Wistar 雄性成年大鼠,体质量 180~210 g,由中国中医科学院中医基础理论研究所实验动物中心提供。实验过程中对动物的处置符合动物伦理学标准。玉红膏购自中国中医科学院广安门医院制剂中心( 批号 20020110) , 京万红为天津达仁堂达二药业有限公司产品( 批号 Z12020440) 。方法: 按完全随机法将大鼠分为玉红膏组、京万红组、模型组,每组 36 只,另设 6 只为正常组。正常组常规饲养不作任何处理,其余各组玉红膏组造模后立即外敷玉红膏,每个创面 0.1 g; 京万红组每个创面外敷京万红0.1 g,每日定时换药 2 次至创面愈合; 模型组创面不予处理。主要观察指标: 于造模后第 1, 3, 5, 7, 14, 21 天 6 个时间点观察记录各组大鼠创面愈合时间并取创面皮缘比较各组各观测点整合素β1 吸光度值及转录因子 p63 阳性细胞数。结果: 纳入大鼠 108 只均进入结果分析。①创面愈合时间: 玉红膏组与京万红组大鼠均短于模型组,差异有统计学意义(P < 0.05) 。②整合素β1 吸光度值及转录因子 p63 表皮层阳性细胞数: 玉红膏组、京万红组及模型组在造模后两指标均高于正常组,差异有统计学意义(P < 0.05); 玉红膏组及京万红组在第 7 天达到峰值,峰值出现的时间早于模型组,峰值的强度高于模型组,差异有统计学意义(P < 0.05)。结论: 玉红膏有促进大鼠创面愈合的作用,该作用可能与其诱导创缘残留的表皮干细胞增殖分化有关。 BACKGROUND: Epidermal stem cells (ESCs) are of importance in the wound repair. Explaining the mechanism that Chinese herb speeds up the regeneration of injured skin from the angle of inducing stem cells deserves to be studied. OBJECTIVE: To observe the effect of Yuhong ointment on the proliferation and differentiation of ESCs during rat wound healing. DESIGN: A randomized controlled animal experiment SETTING : Department of Pharmacology, Wangjing Hospital, China Academy of Chinese Medical Sciences MATERIALS: This study was carried out in the Department of Pharmacology, Wangjing Hospital, China Academy of Chinese Medical Sciences between July and September 2006. Totally 114 healthy Wistar male adult rats, of clean grade, weighing 180-210 g, were provided by the Laboratory Animal Center, Institute for Basic Theory of Traditional Chinese Medicine, China Academy of Chinese Medical Sciences were enrolled in this study. The processing of animals corresponded to the standard of Animal Ethics. Yuhong ointment was purchased from the Pharmaceutical Center, Guanganmen Hospital, China Academy of Chinese Medical Sciences (Lot No. 20020110). Jingwanhong was the product of Tianjin Darentang Da'er Pharmcaceutical Co., Ltd (Lot No. Z12020440). METHODS: The 114 rats were completely randomly chosen and divided into 3 groups with 36 in each: Yuhong ointment group, Jingwanhong group and model group, and the left 6 rats were involved as normal control group. Rats in the normal control group were raised routinely, and no intervention was carried out. Immediately after being modeled, rats in the Yuhong ointment group were spread with Yuhong ointment, 0.1 g each wound, rats in the Jingwanhong group were spread with Jingwanhong, 0.1 g each wound. Dressing change was daily carried out twice regularly until wound healing; The wounds of rats in the model group were untouched. MAIN OUTCOME MEASURES: On the 1^st, 3^rd, 5^th, 7^th, 14^th and 21^th days after modeling, wound healing time of rats in each group was recorded, and integrinβ1 absorbance and transcription factor p63 positive cell amount were compared. RESULTS : Totally 114 rats were involved in the final analysis.① Wound healing time of rats in the Yuhong ointment group and Jingwanhong group was significantly shorter than that in the model group, respectively (P 〈 0.05). ② Integrinβ1 absorbance and transcription factor p63 positive cell amount of modeled rats in the Yuhong ointment group, Jingwanhong group and model group were significantly higher than those in the normal control group, respectively (P 〈 0.05); The above-mentioned two indexes in the Yuhong ointment group and Jingwanhong group reached the peak on the 7m day, which was earlier than peak time in the model group, and peaked intensity of two indexes was significantly higher than that in the model group, separately (P 〈 0.05). CONCLUSION: Yuhong ointment can promote rat wound healing, which may be associated with Yuhong ointment inducing the proliferation and differentiation of ESCS left in the wound edge.
