美洲大蠊提取物对Lewis肺癌小鼠抑瘤作用的研究 被引量:16

摘要 目的探讨美洲大蠊提取物对Lewis肺癌小鼠抑制效应及其作用机制.方法Lewis肺癌小鼠分为生理盐水组、美洲大蠊提取物高、低剂量组,检测各组小鼠的体重变化和抑瘤率,瘤组织HE染色观察病理学变化,应用流式细胞术进行瘤细胞DNA倍体分析.结果美洲大蠊提取物高、低剂量组体重没有生理盐水组增加明显,其抑瘤率分别为41.24%及81.08%;肿瘤组织HE染色发现:美洲大蠊提取物高、低剂量组较NS组瘤细胞坏死增多、瘤组织内及其周围微血管少见;流式细胞术检测美洲大蠊提取物能诱导肿瘤细胞凋亡,随着药物剂量的增高,其凋亡率也升高,并能影响肿瘤的细胞周期,肿瘤细胞S期及G2/M期细胞数大量减少,细胞被阻滞在G0/G1期。结论美洲大蠊提取物对Lewis肺癌小鼠瘤组织的生长具有一定的抑制作用,可促进肿瘤细胞凋亡。 Objective To investigate the inhibitory effect and its mechanism of Periplaneta Americana extract (PAE) on Lewis lung cancer in mice. Methods C57BL/6J mice with 3LL lung cancer, were divided into normal saline (NS), PAE high dose (PAE -H) and PAE low dose (PAE -L) groups. The body weight changes and inhibitory rate of mice in each group were observed. Pathology changes of neoplasia tissue was also observed by HE stain. In addition, the cell cycle and apoptosis index (AI) were analyzed with flow cytometry. Results The body weight increase in PAE -L and PAE -H treated group was less significant than that in NS group and the resistant rates were 41.24% and 81.08%, respectively HE stain indicated that the cell of neoplasia tissue presence nerosis and mocrovessel in PAE high dose (PAE -H) and PAE low dose (PAE -L) groups were fewer than those in NS group. FCM assay indicated that PAE could induce lung cancer cell, with the concentration increase the apoptosis rate became higher. At the same time, the number of S and G2/M phase cells decreased, most of the cells were blocked in G0/GI phase. Conclusion PAE may inhibit the growth 3LL lung cancer in mice and can induce apoptosis of 3 LL lung cancer cell.
