T-2毒素对体外培养大鼠软骨细胞中基质金属蛋白酶13表达的影响 被引量:1

摘要 目的观察T-2毒素对体外培养Wistar大鼠软骨细胞中基质金属蛋白酶13(MMP13)表达的影响。方法培养新生Wistar大鼠关节软骨细胞,按不同质量浓度T-2毒素(0.4、0.8、1.6、3.2μg/L)将实验组分为4组,同时设立对照组(不加T-2毒索)。运用免疫细胞化学方法和免疫印迹方法观察软骨细胞中MMP13的表达水平。结果无论是免疫细胞化学法还是免疫印迹法,各实验组的MMP13表达水平均高于对照组(P<0.01),且随着T-2毒素剂量的逐渐增加,MMP13的表达量也相应增加,各组比较差异均有统计学意义(P<0.01)。结论T-2毒素可增加体外培养软骨细胞中MMP13的表达水平,且呈现剂量依赖关系。这可能是T-2毒素致关节软骨损伤的重要机制之一。 Objective To observe the effects of T-2 toxin on the expression of matrix metalloproteinase 13 (MMP13) in rat chondrocytes in vitro. Methods Isolated and cultured chondrocytes of neonatal Wistar given T-2 toxin (0.4,0.8,1.6,3.2 μg/L, respectively) were assigned into experimental groups, while those free of T-2 toxin were allocated into control groups. After 24 hours, the expression of MMP13 in these chondrocytes were detected with immunocytochemistry and immunoblotting. Results Compared with the control groups, the levels of MMP13 expression in experimental groups increased(P 〈 0.01 ), observed either with immunocytochemistry or immunoblotting. Moreover, the expression of MMP13 in experimental groups was increased gradually along with the increase of the amount of T-2 toxin, the differences among these groups being statistically significant (P 〈 0.01). Conclusions T-2 toxin stimulates chondrocytes in vitro expressing more MMP13 in a dose-dependent manner, which is possibly one of the mechanisms that T-2 toxin leads to damage of articular cartilages.
