西黄丸对人肝癌细胞SMMC7721及小鼠宫颈癌细胞U14周期的影响 被引量:27

摘要 目的研究西黄丸抑瘤作用对肿瘤细胞周期的影响。方法采用体内体外两方面实验进行。体外实验:将西黄丸浸提液加入人原发性肝癌细胞株SMMC7721培养基中,72h后流式细胞术测定SMMC7721细胞的细胞周期;体内实验:连续10d给予移植性U14荷瘤小鼠模型西黄丸,10d后流式细胞术测定U14细胞的细胞周期。结果在体外实验中,西黄丸(200μl/ml)使SMMC7721中G0-G1期细胞比例显著低于肿瘤细胞对照组,G2-M期增多;在体内实验中,西黄丸高剂量组(0.96g/kg)中也导致U14细胞G0-G1期细胞比例显著低于模型组,G2-M期细胞增多。结论体内、体外研究结果显示西黄丸可特异性地将肿瘤细胞阻滞在G2-M期,提示西黄丸可以通过影响肿瘤细胞周期来发挥抑瘤效应。 Objective To study the antitumous effect of Xihuang Pill on cell cycles of cancer cells. Methods Leachate of Xihuang pill was used to treat human malignant tumor cell strains( SMMC7721 )in vitro, after 48 hours the cycle of tumor cells was determined by flow cytometry. At the same time, Xihuang pill was used to treat tumor- bearing mice (U14) for 10 days in vivo, the cycle of U14 cells was determined by flow cytometry. Results Experiment in vitro showed that the proportion of G2 - M of SMMC7721 treated by leachate of Xihuang pill was high (P 〈0.01 ), and experiment in vivo showed that the proportion of G2 - M of U14being treated by Xihuang pill was high too (P 〈0.01 ). Conclusion Xihuang pill can block the tumor cells in G2 -M stage. This indicates that Xihuang pill can affect the cycle of cancer cells so as to block cancer growth.
