摘要
目的筛选土茯苓多糖提取的最优化工艺。方法应用微波辅助提取技术,通过单因素和正交实验,以土茯苓多糖提取率为指标,筛选土茯苓多糖的最佳提取工艺条件。结果土茯苓多糖最佳提取工艺条件为:微波功率800W,固液比(g/ml)1∶30,提取温度100℃,回流浸提两次,10min/次,浓缩液加90%乙醇醇析。结论优化后的工艺稳定,微波辅助提取土茯苓多糖,提取时间短、提取率高,另有较大部分土茯苓总黄酮也同时被浸出。按照优化后工艺条件实验,土茯苓多糖提取率、粗多糖得率以及粗多糖中多糖含量分别为8.33%,3.97%和39.5%。
Objective To optimize the process of extracting polysaccbarides from Smilax glabra Roxb. Methods With the extraction rate of polysaccharides as an examinable target, the optimal extraction conditions of polysaccharides has been proposed by microwave - assisted method through single factor experiment and orthogonal test method. Results The optimal technological parameters of extracting polysaccbarides were as follows: the microwave power, the solid -liquid ratio, the extract temperature and extract time were 800 W, 1: 30(g/ml), 100℃ and 10min twice, respectively. Lastly, the crude polysaccharides were obtained by adding ethyl alcohol into the treated leachate after laying a period. Conclusion The proposed method is stable and reliable for the extraction of polysaccharides from Smilax glabra Roxb. , which has the advantages of short extraction time, high extraction rate and obtaining general flavonoids simultaneously with polysaccharides. Under the optimized operation conditions, the extraction ratio, extraction yield of crude polysaccharides and the content of polysaccharides in the crude polysaccharides were 8.33%, 3.97% and 39.5%, respectively.
