摘要
目的探讨蟾酥对L615白血病的疗效及其可能机制,并对其临床应用提供参考。方法对小鼠行L615白血病造模。造模成功后,蟾酥溶液分别按低、中、高剂量进行腹腔注射给药,1次/d,连续10d,观测小鼠体重变化,生存时间,外周血白细胞计数、分类及血小板计数,肝脾指数,肝、脾、骨髓涂片及骨髓白细胞分类,流式细胞术测定细胞凋亡率。结果L615模型组体重明显减轻,蟾酥低、中、高剂量组体重无明显变化;蟾酥低、中、高剂量组小鼠生存时间比L615模型组长,且中、高剂量具有显著差异(P<0.05或P<0.01);外周血象,同组不同病程,白细胞、血小板计数与实验前比较有显著差异(P<0.05或P<0.01),濒死期各组与L615模型组比较,白细胞、血小板计数,L615白血病细胞百分比有显著差异(P<0.05或P<0.01);白血病死亡小鼠蟾酥各组脾脏指数均低于L615模型组,且有差异(P<0.05);流式细胞术证实,蟾酥能诱导L615白血病细胞凋亡,凋亡率呈剂量依赖性。结论蟾酥对L615白血病具有一定的疗效,其机制之一是诱导白血病细胞凋亡,其次蟾酥能抑制L615白血病细胞对免疫系统的浸润。
Objective To study the therapeutic effects and the potential mechanism of Chansu on the L615 leukemia and provide some advice to its clinical practice. Methods After the model was estabished successfully, Chansu solution （respectively by the low, middle, high dose once a day） was injected intraperitoneally for 10 days continually, the body weight, survival time, peripheral blood leukocyte count and classification, platelet count, hepatosplenic index, hepatic and splenic and bone marrow smears, bone marrow leukocyte classification were observed and the apoptosis rates were determined by FCM. Results The weight in L615 group decreased obviously and the groups injected with Chansu had few changes; the survival time of Chansu groups were longer than those of L615 group, and there were significant differences between middle and high dose Chansu groups （P 〈 0.05 or P 〈 0. 01 ） ; peripheral hemogram, different illness time within the same group, leukocyte count and platelet count changed a lot after the experiment （ P 〈0.05 or P 〈0.01 ） ; during the dying period, compared with the L615 group, the leukocyte and platelet count and the L615 leukemia cell percentage had differences （ P 〈 0.05 or P 〈 0.01 ） ;significant differences was observed between the splenic index of death leukemia mice and those in L615 group （P 〈 0.05 ） ; the FCM proved that Chansu could induce the apoptosis of L615 leukemia cells in dose - dependent manner. Conclusion Chansu has some therapeutic effects to the L615 leukemia. To induce apoptosis is one of its mechanisms. Also, Chansu can inhibit the infiltration of leukemia cells to immune system.
