摘要
目的探索医药用神经节苷脂的最优(纯度)、成本低)生产方法。方法牛脑的氯仿-甲醇提取液通过水草取使神经节苷脂与脂溶性成分分离、再经碱水解除磷脂,上吸附树脂柱,使与水溶性杂质分离。然后作产品鉴定(包括纯度、组成成分、含磷量、活性试验及蛋白反应)。结果所制备的神经节苷脂的纯度为唾液酸含量26%;组成成分:GM116%、GD36%、GD1α/45%、GD1b10%、GT1α14%、GT1b17%;含磷量120μg/100mgGLs;生物活性:较好地促进离体鸡胚背根神经节的存活与生长。结论本法制备的神经节苷脂纯度高、成本低适用于医药用神经节苷脂的生长。
Objective To research an optimal method of preparing ganglioside used in medicine with a good purify and low consumption fo for batch procuce.Methods By means of extraction with water for chloroform- methanol extracte of bovine brain, the gangliosides was isolated from the 1ipid-soluble substances.The phospho1ipids in theextracte solution was remove by aldaline hydrolysis, then the adsorptinn resin column chromatography was carried out to remove the water-soluble substances. The purity,component, phophatide content, biology active of the ganglioside were evaluated.Result The composition of ganglioside prepa-ration was showed by HPTLC are the follows16% GM,, 6% GD,, 45% GD 10% GD,b'14% GT 17 % GT,b' It contains 26% sinllc acid Per 100mg of gangliosides 120μg of phospholipids. The preparation showed a biology activity of promoting survival and groWth of dor-sal root ganglin in vitro.Conclusion This method is suitable to pre-paring ganglioside used in medicine for its good purity and the lowest comsumption.
