毛细管电泳技术分离哺乳动物脑组织中神经节苷脂 被引量:3

Separation of Gangliosides from Mammalian Brain by Capillary Electrophoresis

摘要 利用毛细管电泳(capillaryelectrophoresis,CE)分离猪脑、鼠脑组织中主要几种神经节苷脂GM_1、GD_(1a)、GD_(1b)、GT_(1b),在缓冲液中加入α-环状糊精(α-cyclodextrin,α-CD)来提高分离的选择性和效率,并对缓冲液的pH值、α-CD浓度和电压等变化因素进行研究。结果表明在含15.0mmol/Lα-环状糊精的2.5mmol/L磷酸级冲液(pH8.6)中,电压为25kV时能很好的分离生物样品。从猪脑提取物中分离出GM_3、GM_1、GD_(1a)、GD_(1b)、GT_(1b)和含唾液酸的其他糖脂类物质,鼠脑中分离出GM_1、GD_(1a)和GT_(1b),说明毛细管电泳可用于分离神经节苷脂,具有快速、简便、节约等特点。 Gangliosides are glycosphingolipds containing sialic acid. They have been demonstrated to play important roles in biological functions,such as cell growth,differentiation,malignant transformation, membrane-mediated transfer of information. We have separated several major gangliosides from pig brain and rat brain by capillary electrophoresis (CE). Enhancement of selectivity and efficiency of separation were obtained by using 2. 5 mmol/L phosphate buffer containing 15 mmol/L a-cyclodextrin. We also studied the factors which effect on the separation of the samples. The results indicate that the methods of using capillary electrophoresis can successfully separate gangliosides GM1,GM3,GD1a,GD1b, GT1b from pig brain and GM1,GD1a,GT1b, from rat brain.
