摘要
建立了高效毛细管电泳检测盐酸安妥沙星中对映体杂质的方法。以含20mmol/LHP-β-CD与70mmol/L磷酸二氢钾溶液(用磷酸调节pH3.0)为运行缓冲液,电压30kV,柱温30℃,检测波长297nm。可在10min内将两对映体良好分离,检出限为2mg·L^-1(S/N=3),左旋安妥沙星在4.89—61.2mg·L^-1呈良好的线性关系,日内精密度(RSD)为2.1%(n=5),日间精密度(RSD)为3.2%(n=15)。
Antofloxacin hydrochloride is a new chiral drug. A sensitive, simple and accurate method for enantiomeric impurity determination of antofloxacin hydrochloride was firstly developed by capillary electrophoresis. The optimum experimental conditions were 20 mmol/L HP-β-CD, 70 mmol/L phosphate buffers ( pH 3.0) with an effective voltage of 30 kV at 30 ℃ and detected at 297 nm. The method offers good separation of the two enantiomers in less than 10 min. The limit of detection was 2mg·L^-1(S/N=3). Good linear ranges were obtained from 4.89 to61.2mg·L^-1. The relative standard deviations of intra- and inter-day were 2. 1% (n =5) and 3.2% (n = 15).
