摘要
采用柱前衍生化法同时测定复方α-酮酸片中——L-醋赖氨酸、L-苏氨酸、L-色氨酸、L-组氨酸、L-酪氨酸五种氨基酸的含量。采用ODS柱,以流动相乙腈-水(1:1)和0.14mol/L磷酸盐缓冲液(pH 8.2)梯度沈脱,检测波长360nm。衍生化试剂为2,4-二硝基氯苯。五种氦基酸的平均回收率为99.6%～100.9%,RSD小于1.1%。
A RP-HPLC method with pre-column derivatization was established for simultaneous determination of five amino acids - L-lysine acetate, L-threonine, L-tryptophan, L-histidine, L-tyrosine in compound α-ketoacid tablets. An ODS column was used, with the mobile phase of acetonitrile-water and 0.14mol/L phosphate buffer solution （pH 8.2） by gradient elution, at the detection wavelength of 360nm. 2,4-Dinitrochlorobenzene was used as the derivation reagent. The average recoveries of five amino acids were 99.6 %- 100.9 %, the RSDs were less than 1.1%.
