条件性恐惧对部分脑区即刻早期基因c-fos表达的影响

摘要 目的观察条件性恐惧对大鼠部分脑区即刻早期基因c-fos表达的影响。方法采用条件恐惧训练诱导大鼠焦虑反应模型,免疫组化ABC方法结合计算机图像分析系统对大鼠中枢神经系统海马区和杏仁核区c-fos免疫阳性物质数密度进行测定。结果c-fos免疫阳性物质数密度:中央杏仁核对照组为(0.13±0.13),实验组为(2.71±0.32)(P〈0.01);基底外侧杏仁核对照组为(0.00±0.00),实验组为(1.68±0.25)(P〈0.01);海马对照组为(4.13±0.67),实验组为(3.51±0.97)(P〉0.05)。结论条件恐惧训练后大鼠中央杏仁核和基底外侧杏仁核c-fos表达显著增加,海马区c-fos表达改变不明显。 Objective To investigate the effect of conditioned fear on the expression of the immediate early gene c-fos in special brain regions in rats. Methods Conditioned fear training was used to induce the model of anxiety,immunohistochemical ABC technical and computer assisted image analyzing system were used to detect the optic density of fos-like immunoreactivity (FLI) in amygdala and hippocampus in rats. Results The optic density of FLI in amygdala central :0.13 ±0.13 in control group, 2. 71 ±0. 32 in test group( P〈0, 01 ), The optic density of FLI in amygdale basolateral : 0.00 ±0.00 in control group, 1.68 ±0.25 in test group( P〈0.01 ). the optic density of FLI in hippocampus : 4.13 ± 0.67 in control group, 3.51± 0.97 in test group ( P〉 0.05 ). Conclusion The expression of FLI in amygdala central and amygdale basolateral significantly increased after conditioned fear training,and no significantly change was seen on the expression of FLI in hippocampus.
