摘要
目的:观察海珠益肝胶囊对免疫性肝损伤小鼠神经肽SP、VIP及辅助性T细胞亚群的影响。方法:应用尾静脉注射卡介苗(BCG)和脂多糖(LPS)建立免疫性肝损伤小鼠模型,分离小鼠淋巴细胞,经佛波醇乙酯(PMA)、离子霉素、莫能菌素刺激培养6h后,收获细胞,利用细胞内细胞因子染色流式细胞术检测Th1/Th2比例:采用放免法检测血浆和脑垂体中SP、VIP含量。结果:在模型组,外周血和脑垂体中SP含量较正常组逐渐增加,VIP含量较正常组逐渐降低,第18天、25天与正常组比较有显著性差异(P<0.05);Th1细胞的百分率逐渐升高,Th2细胞的百分率逐渐降低,Th1/Th2的比值增大,与肝组织损害程度呈现正相关:海珠益肝胶囊能够降低外周血和脑垂体中SP含量,增加V lP含量,与模型组第25天比较有显著差异(P<0.01),能够恢复SP与V lP的平衡;降低Th1细胞的百分率,恢复Th1/Th2的比例,对肝组织损害具有保护和改善作用(P<0.05)。结论:在小鼠免疫性肝损伤过程中,存在SP与VIP的分泌紊乱及Th1/Th2细胞失衡,海珠益肝胶囊能够恢复SP/VIP、Th1/Th2细胞的平衡,对肝组织损害具有保护利改善作用,可能是其免疫约理机制之一。
Objective ：To observe the effects of HaiZhu of YiGan Capsule on SP/VIP,Thl/Th2 cells in the peripherl blood mononuclear cells from the mouse with immunological liver injure. Methods ：The ratios of Thl/Th2 cells in the mouse with immunological liver injure were detected by flow cytometry with two color intracellular cytokine staining; the changes in substance P（SP） ,vasoactive intestinal peptide（VIP） in the pituitary gland and peripheral blood were detemi- ned by radio -immunoassary. Results ：In the model group,SP contents in the pituitary gland and peripheral blood gradually increased, and VIP contents gradually decreased, and the changes were significant （ P 〈 0.05 ）. Thl cell ratios were higher, Th2 cell ratios were lower,the ratios of Thl/Th2 were higher,there was positive correlative between the ratio of Thl/Th2 and liver injure. HaiZhu of YiGan Caosule could downregulate the levels of SP. and uoregulate the levels of VIP,and downregulate the radio of Thl cell, and could convert the ratio of Thl/Th2, and could improve live pathological injury（ P 〈 0.05 ）. Conclusion ：There were the disturbance of neuropeptide SP/VIP and Thl/Th2 in the mice with imu- nological hepatic injury, HaiZhu of YiCan Capsule could convert the imbalance of SP/VIP and Thl/Th2, and make it in- cline to the normal state,and make it incline to the normal state,and improve liver pathological injury.
