Effect of Nutrients on Poly(Vinyl) Alcohol Degradation by a Strain of Xanthomonas sp

摘要 基于前期研究分离到的一株能完全降解聚乙烯醇(PVA)的黄单胞菌(Xanthomonas sp),考察了部分简单有机碳源、水溶性维生素、氨基酸对该菌株细胞生长和降解PVA的影响.结果表明,对所考察的3种共代谢基质,0.6 g/L葡萄糖在含2.0 g/L蛋白胨F403条件下,可使1.0g/L PVA完全降解时间缩短至约40 h.0.4 mg/L泛酸钙对细胞生长影响较小,但可促进细胞黄色素的形成,而黄色素含量高的静息细胞对PVA降解量有所增加.20.0 mg/L甲硫氨酸、半胱氨酸可促进细胞明显生长,进而提高PVA降解率. Based on a strain of isolated Xanthomonas sp which could degrade completely Poly (vinyl) alcohol (PVA), the effects of some medium components on cell growth and PVA degradation were investigated, which included several simple carbon sources, aqueous vitamins and amino acids. The results show that glucose is beneficial for PVA degradation among 3 kinds of co-metabolism substrates in the medium with 2.0 g/L peptone F403. The culture time for complete degradation of PVA of 1.0 g/L was shortened to about 40 hours when the concentration of glucose was 0.6 g/L. By the addition of calcium pantothenate of 0.4 mg/L, the growth of bacteria was little influenced, but the production of xanthomonadin in cells was stimulated. The degradation rate of PVA was enhanced for resting cells with more xanthomonadin. By adding methionine or cysteine of 20.0 mg/L, the growth of cells and PVA degradation rate were increased.
