柑橘类果树组织总RNA优异且通用的提取方法 被引量:10

An Effective and General Protocol for the Isolation of Total RNA From the Tissues of Citrus Trees

摘要 分别以柚类、柠檬、甜橙、枳壳和宽皮橘的果皮、果肉、枝皮、老叶、嫩叶、树根、子房、花瓣,尤其是以果胶含量丰富的岩溪晚芦果皮等作试材,将两次LiCl沉淀法加以改良,加入适量CaCl2,找到一种柑橘类果树总RNA优异且通用的提取方法,所得RNA的质量和产量均较高。经差减文库构建、连接、电转化,成功获得了目标片段。 Many samples, such as pulp, epicarp, tender leaves, old leaves, ovary, petal et al were tested respectively, especially the epicarp of Yanxiwanlu fruit very rich in polysacchrides. An effective and general protocol for the isolation of total RNA from the tissues of citrus trees was worked out. The protocol, based on two lithium chloride (LiCl) precipitations, is approved by some steps and added with appropriate CaCl2, during RNA extraction. The results Indicated that the RNA is of high quality, as confirmed by isolation of mRNA and subtraction construction of eDNA libraries.
