摘要
目的:应用流式细胞仪从孕妇外周血中分离胎儿有核红细胞(NRBCs),通过荧光原位杂交(FISH)分析进行无创性产前诊断。方法:对15例妊娠11~24周的高危孕妇外周血按常规密度梯度方法分离单个核细胞,选用红细胞特异性抗体CD71、HbF结合流式细胞术(FACS)分选富集NRBCs,并进行FISH分析。结果:所有标本均分选出NRBCs,分选率为100%,CD71和HbF双阳性细胞含量为(3.43±1.62)×10-2。经FISH成功分析13例,未发现异常结果。结论:从孕妇外周血中分离富集得到的胎儿有核红细胞,通过FISH分析检测胎儿特定的染色体,可进行无创性产前诊断遗传性疾病。
Objective: To separate fetal nucleated red blood cells from the peripheral blood of pregnant women with FACS and FISH is feasible for noninvasive prenatal diagnosis of genetic diseases. Methods: Mononuclear cells in peripheral blood samples from 15 pregnant women from 11 to 24 weeks of gestation we:e isolated useing density gradient centrifugation. NRBCs were sorted with fluorescene - activated cell sorting (FACS) by erythrocyte specific antibody CD71 and fetal specific antibody HbF. Fluorescenece in situ hybridization (FISH) was performed. Results: Fetal NRBCs were separated from all cases and the content of CD71 and HbF double -positive cells was (3. 43 ± 1. 62) ×10^ -2. 13 cases were performed by FISH and abnormality was not detected. Conclusion: Te method that isolating and enriching fetal NRBCs from the peripheral blood of pregnant women and detecting special chromosomes of foetus by FISH is used to diagnose genetic diseases.
