Analysis of Chlorogenic Acid and Rutin in the Flue-cured Tobacco Leaves by High Performance Liquid Chromatography

摘要 探索了高效液相色谱法分离绿原酸和芸香苷的试验条件。研究结果表明,采用沸程60~90℃石油醚抽提,用磷酸二氢钾和甲醇溶液作为流动相进行梯度洗脱,可以较好地分离测定烤烟叶中绿原酸和芸香苷的含量。该法不仅具有良好的线性(相关系数为0.9984~0.9986)和重复性(相对偏差小于3.11%),而且绿原酸和芸香苷的平均回收率可达到97.99%和97.29%。对国内外烤烟叶分析的结果表明,湖南郴州、巴西和赞比亚烟叶中绿原酸含量较高;美国烟叶中的芸香苷含量较低;南非烤烟绿原酸和芸香苷含量的比值最低。 A method for separation and determination of chlorogenic acid and rutin in flue-cured tobacco leaves by high performance liquid chromatography(HPLC) was developed. The sample was extracted by petroleum ethers whose boiling point ranged from 60 -90 ℃, and the mobile phase was potassium dihydrogen phosphate buffer solution and methanol on gradient elution, which make to violate and measure the chlorogenic acid and rutin. The results showed this method has good linearity with correlation coefficient 0. 998 4 to 0. 998 6, and relative standard deviation was less than 3.11%. The recovery on average was 97.99% and 97.29% respectively. The analyse results of flue-cured tobacco samples from the internal and external countries showed that the content of chlorogenic acid in Chenzhou ( Hunan), Brazil and Zambia is higher than that in the other countries. The rutin content of American tobacco is lower than the others. The ratio of chlorogenic acid to rutin in tobacco of South Africa is the lowest.
