高效液相色谱法同时测定枸杞中单糖和低聚糖的含量 被引量:16

Simultaneous Determination of Monosaccharides and Oligosaccharides in Lycium barbarum L. by High Performance Liquid Chromatography

摘要 目的建立同时测定枸杞中鼠李糖、果糖、葡萄糖三种单糖和蔗糖、麦芽糖两种低聚糖含量的高效液相色谱一示差折光检测方法。方法枸杞中的单糖和低聚糖用蒸馏水提取后,经Zorbax Carbohydrate色谱柱分离,采用示差折光检测器检测。结果本方法检测枸杞中的鼠李糖、果糖、葡萄糖、蔗糖、麦芽糖在10.0~400.0mg/L范围内,质量浓度与色谱峰面积呈良好的线性关系,相关系数r=0.9976~0.9998。方法的精密度RSD分别为2.15%、1.49%、1.09%、3.82%、4.65%,加标回收率为92.27%~101.93%,方法的检出限(信噪比S/N=3)分别为4mg/kg、5mg/kg、4mg/kg、6mg/kg、5mg/kg。结论高效液相色谱一示差折光检测方法简便、快速,适用于枸杞中鼠李糖、果糖、葡萄糖、蔗糖、麦芽糖含量的测定。 Objective To develop a rapid approach of refractive index detection with high performance liquid chromatography (HPLC) for the determination of rhamnose, fructose, glucose, surose and maltose in Lycium barbarum L. Methods The sample was extracted with water and the analyte was separated by Zorbax Carbohydrate column with aeetonitrile-water as mobile phase. Results Good linear correlations between the concentrations and the peak areas of the analyte were found, with the correlation coefficients ranging from 0. 9976 to 0. 9998. The spiked recovery rates ranged from 92.27% to 101.93%, with 1.85%-3.27% of relative standard deviations (n~ 5). The limits of detection (S/N= 3) were 4-6 mg/kg. Conclusion The proposed method is suitable for the determination of monosaccharide and oligosaccharide in Lycium barbarum L.
