摘要 目的探讨Vilon(L-Lys-L-Glu)对参与炎症反应的小鼠腹腔巨噬细胞分泌IL-1β、NO的影响。方法以小鼠原代培养腹腔巨噬细胞为阳性参照,ELISA法检测Vilon及脂多糖(LPS)共刺激的小鼠腹腔巨噬细胞IL-1β的分泌水平;还原酶法分析NO分泌水平;RT-PCR法检测IL-1β和iNOS mRNA的表达。结果Vilon和LPS共刺激小鼠腹腔巨噬细胞时,Vilon对LPS活化的小鼠腹腔巨噬细胞分泌IL-1β及NO具有明显的促进作用,并且呈剂量依赖关系;同时也促进了IL-1β和iNOS mRNA表达。结论Vilon对活化的小鼠腹腔巨噬细胞分泌IL-1β、NO具有明显的促进作用。 Objective To investigate the effects of Vilon(L-Lys-L-Glu)on IL-1β and NO secretion of murine peritoneal macrophages. Methods Taking LPS-activated murine peritoneal macrophages as positive controls, the level of IL-1β secreted by murine peritoneal macrophages co-stimulated with Vilon and LPS was examined by ELISA, the level of NO was detected by NO analyzing Kit, and the expressions of IL-1β mRNA and iNOS mRNA were analyzed by RT-PCR. Results Vilon promoted the secretion of IL-1β and NO, meanwhile the express/on level of IL-1β mRNA and iNOS mRNA were increased by Vilon in the LPS-activated routine peritoneal macrophages. Conclusion Vilon increases the secretion of IL-1β and NO of murine peritoneal macrophages activated by LPS.
