摘要
目的:研究高能X线照射后不同时间点人肝癌细胞株HepG2凋亡及bcl-2、Fas表达的变化,为原发性肝癌放射治疗的时间剂量分割提供借鉴。方法:用吸收剂量为5Gy的6MVX线单次照射HepG2细胞,以流式细胞仪测定受照后不同时间的细胞凋亡率,以免疫细胞化学法检测细胞受照前后bcl-2和Fas的表达水平。结果:高能X线可诱导HepG2细胞发生凋亡,照射后6h细胞凋亡率达到高峰(32%),同时bcl-2表达水平下降,Fas表达水平升高。结论:高能X线能够诱导肝癌细胞株HepG2发生凋亡,bcl-2及Fas可能参与其细胞凋亡的调控。
Objective: To study the relationships between time course and apoptosis as well as bcl-2 and Fas expressions in human hepatocarcinoma cell line HepG2 exposed to high-energy X-ray in vitro. Methods: HepG2 cells were exposed to 6MV X-ray with a single dose of 5 Gy. Apoptosis was detected with PI method by flow cytometry at different time points after the exposure. The expression of bcl-2 and Fas were detected by immunocytochemistry at different time points before and after the exposure. Results: Apoptosis was induced by high-energy X-ray in human hepatocarcinoma cell line HepG2.The apoptosis rate of HepG2 cells reached its peak (32%)at 6 hrs after the irradiation. The bcl-2 expression decreased and Fas expression increased in the course of apoptosis induced by high-energy X-ray in HepG2 cell line. Conclusion: Apoptosis was induced by high-energy X-ray in HepG2. bcl-2 and Fas might play a role in apoptosis induced by X-ray in HepG2 cells.
