摘要
确定了以啤酒废酵母为原料,酵母酶解液中α-氨基氮含量为考察指标的蜗牛酶酶解破壁条件;并以啤酒废酵母酶解液为有机氮源,厌氧发酵制备丁二酸。结果表明蜗牛酶破壁效果最好,其最佳反应条件为蜗牛酶用量10mg/g干物质,pH6.5,40℃水浴,反应16h,酵母酶解液中α-氨基氮含量为0.504g/100mL;当发酵培养基中葡萄糖浓度为50g/L,啤酒废酵母酶解液作为有机氮源时,菌株Actinobacillus succinogenes NJ113可以产34.6g/L丁二酸,收率69%,和以酵母膏为有机氮源厌氧发酵制备丁二酸(丁二酸浓度为35.1g/L,收率70.2%)比较,产量相近,成本较低。
Yeast cell rupture conditions by zymolysis were determined with waste brewer's yeast as material and α-amino nitrogen as inspection target. The research on the anaerobic production of succinic acid by using waste brewer's yeast solution as organic nitrogen source was studied. The results showed glusulase was the best rupture enzyme and its optimum reaction conditions were glusulase 10mg/g dry matter, pH6.5, 40℃, 16h, the concentration of α-amino nitrogen was 0.504g/100mL. Actinobacillus succinogenes NJl13 could produce 34.6g/L succinic acid with the yield of 69% from 50g/L glucose and waste brewer's yeast solution as organic nitrogen. Compared with the production of succinic acid by using yeast extract as organic nitrogen source (succinic acid 35.1g/L, yield 70.2%), the concentration was near and cost is lower.
