摘要
目的观察地龙蛋白组分Ⅲ对鼻咽癌细胞裸鼠移植瘤血管密度和MMP-9表达的影响。方法裸鼠接种HNE1细胞形成移植瘤,至肿瘤体积达1cm×1cm×1cm后,随机分为地龙组和对照组,分别于腹腔注射地龙蛋白组分Ⅲ和生理盐水,连续用药20天,然后处死动物,测量鼻咽癌细胞裸鼠移植瘤的体积重量,并制备组织切片,显微镜下计数瘤旁血管密度,免疫组化S-P法检测组织中金属基质蛋白酶9的表达活性。结果地龙组用药处理后,与对照组进行比较,地龙组裸鼠移植瘤的体积和重量均明显降低(P<0.05),瘤旁血管密度明显减少(P<0.05),金属基质蛋白酶9表达明显减弱(P<0.05)。结论地龙蛋白组分Ⅲ可能通过抑制金属基质蛋白酶9的表达活性和抑制瘤旁微血管的生长而抑制鼻咽癌细胞的转移潜能。
Obuective To investigate the effects of component protein Ⅲ contracted from Pheretima aspergillum on the microvessel density and expressing activity of matrix metalloproteinase 9 mRNA in the implanted tumors with nasopharyngeal carcinoma(NPC) cells among nude mice. Methods NPC cell line HNE1 was used to prepare the implanted tumors among nude mice by injection methods a first,and then, these animals, with the implanted tumors grown to a volume about 1cm × 1 cm × 1 cm, were randomly divided into two groups, with 10 mice in each group. The mice of experimental group were injected intraperitoneal injection with the component protein Ⅲ of Pheretima aspergillum and those of controlling one were injected with the sane volume of nomal saline in the same way,all lasted for 20 days. Thereafter, all the animals were put into death and their tumors were removed to prepare sections for microvessel density and expressing activity of matrix metalloproteninases 9 mRNA detemining by optical microscopy and immunohistochemical S-P procedures respectively following the volumes and weights of implanting tumors measured. At last, a comparative analysis was made to see the differences of measured indicators between the two groups to evaluate the implications of such a greating procedure in this study. Results Following the treatment with the component protein Ⅲ of Pheretima aspergillum for 20 days, the measured volumes and weights of implanted tumors were boths hown significantly reduced among the animals of experimental group, when compared with of controlling one(P 〈 0.05 ), with significantly decreased para-tumor microvessel density ( P 〈 0.05 ) and weakened expressive activity of matrix metalloproteinase 9 mRNA in the tumor tissues(P 〈 0. 05). Conclusion It should be reasonable that the component protein Ⅲ of Pheretima aspergillum can bring about inhibitory effects on the metastatic potentiality of NPC cells by blocking growth of new microvessels in the tissuse rounding the tumor and by inhibiting the expressive activity of matrix metalloproteinases 9 mRNA in the tumor tissues.
