乳腺癌患者放疗前后外周血淋巴细胞HPRT基因位点突变频率的初步研究 被引量:1

摘要 目的:研究乳腺癌患者手术及放疗前后外周血淋巴细胞HPRT基因突变频率的变化,探讨体细胞HPRT基因突变频率与乳腺癌手术及放疗的关系。方法:采用多核细胞法研究乳腺癌患者手术前后及放疗前后外周血淋巴细胞HPRT基因位点的突变频率,并与正常人群进行对比。结果:去除吸烟以及年龄对HPRT基因突变频率的影响后,乳腺癌患者手术前的外周血淋巴细胞HPRT基因突变频率均数为1.118×10-3,高于对照组均数1.040×10-3,t=6.848,P<0.01;乳腺癌患者手术后HPRT突变频率均数1.103×10-3,与手术前HPRT突变频率均数1.118×10-3比较,差异无统计学意义,t=1.328,P>0.05。乳腺癌患者放疗后外周血淋巴细胞HPRT基因突变频率均数为1.187×10-3,高于放疗前均数1.103×10-3,t=7.179,P<0.01。结论:HPRT基因位点突变频率可能适合乳腺癌高危人群的筛选和生物学检测,也有可能作为放疗损伤的生物学监测指标。 OBJECTIVE: To study the HPRT locus mutation frequency in peripheral blood lymphocytes in breast cancer patients before and after both surgery and radiotherapy, and to explore the relationship between surgery and radiotherapy. METHODS: Multinuclear cell assay was used to study the frequency of HPRT locus mutation in breast cancer patient's before and after both surgery and radiotherapy, and the results were compared with the normal people. RESULTS: After getting rid of the factors such as age and smoking which may impact the frequency of HPRT locus mutation, the mean frequency of HPRT locus mutation in breast cancer patients before surgery ( 1.118 × 10^-3 ) was significantly higher than that in healthy people (1. 040 × 10^-3 ), t=6. 848, P〈0.01. There was no significant difference after surgery compared with that before surgery, but the mean freqvency of HPRT locus mutation after radiotherapy (1. 187 × 10^-3) was significantly higher than that before radiotherapy ( 1. 103 × 10^-3 ), t = 7. 179,P〈0. 01. CONCLUSION: The HPRT locus mutation in peripheral blood lymphocytes may be useful for screening and biomonitoring those breast cancer susceptible people, and expected to be a biomarker for radiotherapy injury assessment.
