摘要
目的观察维甲酸制作的大鼠骨质疏松模型中,肝脏内铁调素(Hepcidin)基因在不同时间表达变化,探讨其与骨质疏松形成的相关性。方法取80只SD大鼠,分成实验组和对照组。实验组用维甲酸(70mg/kg.d)每天灌胃,14d后形成骨质疏松模型;在实验组中采用RT-PCR方法分别观察灌胃第1、3、5、8、9、10、12、14d肝脏铁调素基因表达变化;在第14d还检测大鼠骨密度、分析脱钙骨切片、观察肝组织切片。结果与对照组相比,实验组大鼠肝脏铁调素基因表达在维甲酸灌胃的不同时间组存在明显不同:早期铁调素相对表达量逐渐下降,在第8、9d达到最低值,随后有回升。另外,维甲酸灌胃第14d的骨密度比灌胃前明显降低、骨切片中骨小梁稀松皮质骨变薄;维甲酸灌胃第14d的肝组织切片与对照组比较差异无显著性。结论①本研究骨质疏松模型采用人工干预方法,没有其他影响因素。实验结束时大鼠骨密度、骨组织学检查表明骨质疏松形成;因此,同期不同时间组大鼠体内铁调素基因表达可以代表体内铁代谢不同时间的变化。②在本研究中实验组与对照组研究结果的差异表明:铁调素基因RT-PCR变化与维甲酸制作的大鼠骨质疏松模型形成有实验相关性。
Objective To observe changes of hepatic hepcidin gene expression on different stage in experimental osteoporosis model induced by retinoic acid and to investigate associations between hepcidin and osteoporosis. Methods 80 SD rats were randomly divided into test group and control group; osteoporosis model group rats were administered with retinoic acid(70 mg/kg· d) continuing 14 days by intragastric administration. RT-PCR was applied to detect changes of hepatic hepcidin gene expression on model different stage. Bone density was detected and decalcification bone micmtome section and hepatic histological section were observed and analyzed on the 14th day. Results Compared with control group, hepatic hepcidin gene expression of test group on different stage had obvious differences. On early stage of test group, relative contents of hepcidin gradually fell and the lowest level was on 8th and 9th day, then the hepcidin level gradually rose. In addition, rats in test group on 14th day, bone density was evidently lower, bone trabecula of decalcification bone microtome section were looser and os integumentale were thinner than when retinoic acid was not administered. Meanwhile hepatic tissue microtome section had no significant deviation between two groups. Conclusions 1. The osteoporosis model of our study applied man-made interferences, there was no other influential factors. On the end stage of study, bone density and bone histological anatomy indicated that osteoporosis model had formed. Therefore, internal hepcidin gene content of rats that were in different time groups of synchronization can represent changes of iron metabolism on different time stages. 2. In the study, differences between test group and control group indicat that there is association between change of hepcidin gene level and osteoporosis model induced by retinoic acid.
