蜕膜细胞条件培养液中TNF-α及EGF对卵巢癌细胞侵袭基因uPA/PAI-1表达的影响

摘要 目的研究蜕膜细胞条件培养液(decidual cell conditioned media,DCM)中肿瘤坏死因子(TNF-α)及表皮生长因子(EGF)对卵巢癌细胞株(COC1)侵袭基因尿激酶型纤溶酶原激活因子(uPA)/纤溶酶原激活物抑制因子1(PAI-1)表达的影响。方法原代培养早孕蜕膜细胞及晚孕蜕膜细胞并提取蜕膜细胞条件培养液(DCM),分别将加有TNF抗体及EGF抗体的DCM处理卵巢癌细胞株(COC1),利用RT-PCR法对卵巢癌侵袭基因uPA/PAI-1的表达进行分析。结果COC1表达PAI-1,不表达uPA。与DCM组比较,TNF-α抗体组可以使COC1PAI-1mRNA的表达上调,二者之间差异有显著性(P<0.01)。与DCM组比较,EGF抗体组PAI-1mRNA的表达不明显,呈下调的趋势,二者之间亦差异有显著性(P<0.01)。结论早孕DCM中TNF-α降低了DCM抑制COC1纤溶酶原系统作用的能力;而EGF增加了DCM的上述能力。同时说明纤溶酶原系统在COC1的侵袭能力方面不起主导作用。
