低氧环境对大鼠血清bFGF水平的影响及沙利度胺的干预效应 被引量:4

摘要 目的观察低氧环境对大鼠血清bFGF水平的影响,并探讨低氧条件下血管生长抑制剂沙利度胺对血清bFGF水平的干预效应。方法采用低压氧舱模拟4000m海拔的高原低压低氧环境。将实验动物分为低氧组、沙利度胺预防组和沙利度胺治疗组,经不同时间的低氧处理及沙利度胺干预后,测定血清bFGF水平,并与饲养在西宁地区(海拔2260m)自然条件下的对照组大鼠进行比较分析。血清bFGF的测定采用固相双抗体夹心酶联免疫吸附实验(ELISA)方法测定。结果低氧组大鼠血清bFGF浓度呈现随低氧时间的延长而不断上升趋势(对照组:7.38±1.0pg/ml;低氧1天组:7.47±1.86pg/ml;低氧7天组:16.86±1.91pg/ml;低氧21天组:35.92±23.52pg/ml),低氧不同时间组间及低氧7天组和低氧21天组与对照组间血清bFGF浓度存在显著差异(P<0.01)。经沙利度胺进行预防性干预,未能阻断低氧引起的血清bFGF水平增高,但显著降低了增高幅度(低氧预防7天组:12.28±1.87pg/ml,低氧预防21天组:19.26±2.67pg/ml),预防7天组血清bFGF水平与对照组比较有升高(P<0.05),但显著低于低氧7天组(P<0.05);预防21天组血清bFGF水平比预防7天组显著增高,却显著低于低氧21天组(P<0.05)。经沙利度胺治疗后,治疗组血清bFGF水平恢复到与对照组无差异的水平(P>0.05)。结论在低氧环境中,大鼠血清bFGF浓度呈现随低氧时间的延长而不断增高的趋势;沙利度胺可部分阻止低氧引起的大鼠血清bFGF的增高;沙利度胺对低氧引起的血清bFGF水平增高具有治疗作用。沙利度胺在低氧引起的血管增生性疾病的预防和治疗中可能具有一定作用。 Objective observe the serum levels of Basic Fibroblast Growth Factor (bFGF) in the hypoxic environment and to reveal the interferential effects of blood growth inhibitors - thalidomide on serum bFGF in the same condition. Methods By means of low pressure oxygen chamber, the low pressure hypoxic environment at 4000 - meter -high plateau was simulated. Experimental animals were divided into three groups as the hypoxic group, the preventive group with stopped reaction and the treated group by using thalidomide. After various periods of hypoxic treatment and intervention thalidomide, Serum bFGF was measured and compared with control group which wasraised in the natural environment in Xining 2260 meters high. Serum bFGF was detected by using solid - phase double - antibody sandwich enzyme - linked immunosorbent assay (ELISA) Results The concentrations of the serum bFGF of the rats in hypoxic group increased with the extension of hypoxic time ( the control group : 7.38 ± 1.0pg/ml, one - day hypoxic group :7.47 ± 1.86pg/ml. 7 - day - hypoxic group : 16.86 ± 1.91 pg/ml, 21 - day -hypoxic group: 35.92 ± 23.52 pg/ml respectively). Significant differences were found among those groups(P 〈 0.01 )even though preventive intervention implemented by thalidomide could not block the increase of serum bFGF level, it decreased the raising extent with big margin. (7 -day -hypoxia preventive group: 12.28 ± 1.87pg/ ml, 21 -day- hypoxia preventive group: 19.26 ± 2.67pg/ml respectively). Compared with the control group, the serum bFGF levels of 7 - day - preventive group increased not a little ( P 〈 0.05 ) , but were much lower than that of 7 - day - hypoxic group. Meanwhile, compared with 7 - day - preventive group, the serum bFGF levels of 21 - day - preventive group increased significantly and were much lower than that of 21 - day - hypoxic group(P 〈 0.05). After thalidomide treatment, the serum bFGF levels of treatment group returned to that of the control group without much difference. (7.28 ± 1.56 pg/ml) ( P 〉 0.05 ) Conclusion The results above show that the concentrations of the serum bFGF of the rats increase with the extension of hypoxic time. Thalidomide, in part, could block the increase of serum bFGF level. Thalidomide has treatment effect on the decrease of the serum bFGF level caused by hypoxic environment. For vascular proliferating disease caused by hypoxic environment, thalidomide plays a certain role in the prevention and treatment of the disease to some extent.
