摘要
用自旋捕集和电子自旋共振技术及化学发光法观察了雷公藤二萜类成分内酯醇(T(10))和雷酚内酯(D7)对水溶系统与细胞体系氧自由基的影响。结果提示在黄嘌呤-黄嘌呤氧化酶系统中,T(10)和D7对O2信号表现明显抑制而不抑制黄瞟呤氧化酶活性。在Fenton反应系统,T(10)对OH无清除作用D7则能部分清除OH。T(10)μg/ml、D7,2μg/ml和3μg/ml时能抑制人多核白细胞的化学发光。且不影响其氧耗,但T(10)在3μg/ml则使多核白细胞的化学发光增强。结果证实,二种成分均有一定清除氧自由基活性的能力。
The scavenging effects of tripto1ide (T(10)) and another diterpene lactone epoxide (D7) extracted from Tripterygium wilfordii on oxygen free radicals (OFRs) were detected by electron spin resonance (ESR)and spin trapping, and by chemiluminescence (CL) methods- Both T(10). and D7 scavenged superoxide anion radical (O2) effectively in xanthine-xanthinoxidase system, and there was no effect on the activity of xanthinoxidase. T(10) didn't scavenge hydroxyl radical (OH), whereas D7 did partially at the concentration of 5μg/ml in fenton reaction system. CL of human polymorphonuclear leukocytes stimulated by phorbol myristate was inhibited by T(10) at 1μg/ml and D7, at 2μg/ml concentration and there was no effect on oxygen consumption in the course. These results suggest that T(10) and D, have the capacity of scavenging OFRs.
